Título : PAPEL FUNCIONAL DEL β-DISTROGLICANO DURANTE LA GRANULOPOYESIS

Resumen : Hematopoiesis is the process in which multipotential hematopoyetic stem cell is self-renewal and differentiates giving place to all the cellular lineages of blood. Cellular and adhesive interactions between hematopoietic stem cells and its microenvironment represent a fundamental role in the regulation of hematopoiesis in the level of cellular proliferation and differentiation. The blood tissue is composed of red blood cells, white blood cells and platelets. According to its morphology and function white blood cells are grouped into three categories: granulocytes, monocytes and lymphocytes. Neutrophils constitute the most numerous kind of granulocytes taking part in the inflammatory process and fagocytosis. Dystroglycan is an adhesion cellular glycoprotein with ubiquitous distribution, it forms part of a complex of proteins, the DAPC (Dystrophin-associated protein complex), associating in a similar way with utrophin. DAPC has been mainly characterized in muscular and nervous tissue, being widely studied in neuromuscular disorders. In blood tissue a long term study has been started to evaluate the expression and distribution of this proteins in progenitors and mature cells. In the present project a study of the role of dystroglycan during granulopoiesis was realized, using HL60 cells of acute promyelocytic leukemia as a model. These cells are induced to granulopoiesis with the aid of dimethyl sulfoxide (DMSO). The presence of the pattern expression of dystrophin and utrophin as well as the proteins of DAPC, giving emphasis to β-dystroglycan was characterized. It was determined that in HL60 cells β-dystroglycan had a molecular weight of proximately 31kDa, smaller than the expected of 43kDa, after induction of granulopoiesis process, the same band of 31kDa was observed. However, the presence of another band of 52kDa proximately was observed. Apparently the presence of the lower molecular weight band to that expected is the product of the molecular full weight protein degradation by extracellular matrix metalloproteinases. Our results strongly suggest that metalloproteinase 9 plays an important role in such degradation. An important function of the molecular full weight protein, β-dystroglycan during the processes of differentiation in these cells is discussed.
Descripción : La hematopoyesis es el proceso en el cual una célula madre hematopoyética pluripotencial se auto-renueva y diferencia dando lugar a todos los linajes celulares sanguíneos. Las interacciones celulares y adhesivas entre células madre hematopoyéticas y su microambiente representan un papel fundamental en la regulación de la hematopoyesis, a nivel de proliferación y diferenciación celular. El tejido sanguíneo está conformado por glóbulos rojos, glóbulos blancos y plaquetas. Con base a su morfología y función los glóbulos blancos se agrupan en tres categorías: granulocitos, monocitos y linfocitos. Los neutrófilos constituyen el tipo de granulocitos más numerosos participando en los procesos inflamatorios y de fagocitosis. El distroglicano es una glicoproteína de adhesión celular con distribución ubicua, forma parte del DAPC (complejo de proteínas asociadas a distrofina), pudiendo asociarse de modo similar con Utrofina. El DAPC ha sido caracterizado en tejido muscular y nervioso principalmente, siendo ampliamente estudiado en los desordenes neuromusculares. En tejido sanguíneo se ha iniciado un estudio a largo plazo para evaluar la expresión y distribución de estas proteínas en componentes diferenciados y progenitores. En el presente proyecto se realizó un estudio del papel del distroglicano durante la granulopoyesis, utilizando como modelo células HL60, de leucemia promielocitica aguda, dichas células son inducibles a granulopoyesis con ayuda de dimetil sulfoxido (DMSO). Se caracterizó la presencia del patrón de expresión de distrofina y utrofina, así como del resto de las proteínas del DAPC, poniendo énfasis en β distroglicano. Se determinó que en las células HL60 β-distroglicano tiene un peso molecular de 31 kDa, menor al esperado de 43 kDa, tras la inducción al proceso de granulopoyesis se aprecia la misma banda de 31 kDa, sin embargo se observa la presencia de otra banda de aproximadamente 52 kDa. La presencia de una banda de menor peso molecular al esperado puede ser debido a la degradación de la proteína de peso molecular completo por metaloproteasas de matriz extracelular, y al parecer, la metaloproteasa 9 tiene un papel importante en dicha degradación. Se sugiere un papel importante de la proteína de peso molecular completo, β distroglicano, durante los procesos de diferenciación en estas células
