Resumen : In the field of transfusion medicine, it is important to study and determination of the Duffy blood group and Diego in donors and in patients who undergo transfusions or transplants. In the anthropological field are considered as markers of population or genetic identity. There have been some studies regarding the phenotype of the Duffy and Diego systems in the Mexican population, however, to date have not been characterized genotypes to determine the correlation between the two studies and use them as a tool in clinical studies and populations. Objective: To determine the major alleles of the Duffy and Diego blood groups in a sample of the resident population of the Mexico City metropolitan area. Methods: DNA samples were obtained from donors at Medica Sur Hospital and INPer. We used real-time PCR and allele specific to characterize the alleles FY * A, FY * B, FY * X, FY * NULL, DI * DI * B AY using the LightCycler ®. Hybridization probes were used LightCycler FastStart DNA Master HybProbe and EZ Type ® Red Cell for the determinations. Results: We studied 403 individuals for the Duffy blood group, frequency phenotypic found to Fya + b-, Fya + b + Fya-b + and Fya-b-was 36.7, 43.5, 17.1 and 2.7% respectively, while for alleles FY * A, FY * B, FY * 0 and FY * X frequencies were 0.543, 0.419, 0.026 and 0.012, respectively. concordance between genotype and phenotype for the Duffy system, where this was 0959, for phenotypes Fya + b + and Fya-b + was 0.971 in both cases, Fya + b-0912 and the null phenotype (Fya-b-) was 0.546, which was in the group that found the majority of discrepancies. The system fenotipicación Diego and genotyping was performed in 247 subjects, detected phenotypes and Day + b + a-b + Di in 17.8 and 82.2% respectively, did not identify individuals with phenotype Day + b-. The allele frequency for the DI * A was 0.085 and 0.915 DI * B, not found DI * A allele in homozygous form in any donor. As the overall agreement for this system of 0972 and 0909 phenotype for Day + b + and 0990 for Dia-b +. Conclusions: For the population studied Duffy system is more like whites than blacks by the higher frequency of allele FY * A. The predominant mechanism for Duffy null phenotype in our population, the Philippines (FY * 0 on FY * A allele). The Dia antigen of the Diego system, is found in very few individuals at a marker that is considered dangerous in terms of population and transfusion.
Descripción : En el campo de la medicina transfusional, es importante el estudio y la determinación de los grupos sanguíneos Duffy y Diego en los donadores y en los pacientes que se someten a transfusiones o trasplantes. En el ámbito antropológico son considerados como marcadores de identidad poblacional o genética. Se han realizado algunos estudios en relación al fenotipo de los sistemas Duffy y Diego en población mexicana, sin embargo, a la fecha no se han caracterizado los genotipos con el fin de determinar la correlación existente entre ambos estudios y poder usarlos como herramienta en estudios clínicos y de poblaciones. Objetivo: Determinar los alelos principales de los grupos sanguíneos Duffy y Diego en una muestra de la población residente de la Ciudad de México y área conurbada. Material y métodos: Las muestras de ADN se obtuvieron de donadores del Hospital Médica Sur y del Instituto Nacional de Perinatología, SS.. Se usó la PCR en tiempo real utilizando el LightCycler®. con las sondas de hibridación LightCycler FastStart DNA Master HybProbe y la PCR alelo específica Red Cell EZ Type ® para caracterizar los alelos FY*A, FY*B, FY*X, FY*NULL, DI*A Y DI*B. Resultados: Se estudiaron 403 individuos para el grupo sanguíneo Duffy, la frecuencia fenotípica encontrada para Fya+b-, Fya+b+, Fya-b+ y Fya-b- fue de 36.7, 43.5, 17.1 y 2.7 %, respectivamente, mientras que para los alelos FY*A, FY*B, FY*0 y FY*X las frecuencias fueron 0.543, 0.419, 0.026 y 0.012, respectivamente .La concordancia global entre el genotipo y fenotipo para el sistema Duffy, donde esta fue de 0.959; para los fenotipos Fya+b+ y Fya-b+ fue de 0.971 en ambos casos, Fya+b- 0.912 y para el fenotipo nulo (Fya-b-) fue de 0.546, que fue en el grupo que se encontró la mayoría de discrepancias. La fenotipicación y genotipificación del sistema Diego se realizó en 247 sujetos, detectándose los fenotipos Dia+b+ y Di a-b+ en 17.8 y 82.2% respectivamente, no se identificaron individuos con fenotipo Dia+b-. La frecuencia para el alelo DI*A fue de 0.085 y para DI*B de 0.915, no se encontró el alelo DI*A en forma homocigota en ningún donador. Siendo la concordancia global para este sistema de 0.972 y por fenotipo de 0.909 para Dia+b+ y de 0.990 para Dia-b+.La concordancia global entre el genotipo y fenotipo para el sistema Duffy, donde esta fue de 0.959; para los fenotipos Fya+b+ y Fya-b+ fue de 0.971 en ambos casos, Fya+b- 0.912 y para el fenotipo nulo (Fya-b-) fue de 0.546, que fue en el grupo que se encontró la mayoría de discrepancias. Siendo la concordancia global para este sistema Diego de 0.972 y por fenotipo de 0.909 para Dia+b+ y de 0.990 para Dia-b+. Conclusiones: Para el sistema Duffy la población estudiada es más parecida a los caucásicos que a los individuos de origen negroafricano, por la mayor frecuencia del alelo FY*A. El mecanismo predominante para el fenotipo Duffy nulo en nuestra población, es el filipino (FY*0 sobre el alelo FY*A). El antígeno Dia del sistema Diego, se encuentra en muy pocos individuos por lo que se considera un marcador poblacional y peligroso en términos transfusionales
