Título : ESTUDIO DEL EFECTO DEL CAMPO ELÉCTRICO SOBRE LA ISOMERÍA DE LOS ÁCIDOS GRASOS DEL AGUACATE

Resumen : Avocado and avocado products are having increased demand over the last decades, however avocado has a very short shelf life and new technologies are under current research. The main challenge is decrease the activity of the polyphenoxidase (PPO) enzyme, which causes brown color in the avocado pulp. In the present work the effect of a electric field (EF) on the fatty acids of the avocado-oil was evaluated, when the EF is applied to diminish the activity of the PPO. It is well known that the avocado oil contain an important amount of functional fatty acids which can be affected by the use of the EF. Some related articles indicate that the extraction method of the oil could be a factor that increases the trans fatty acids, (TFA). The idea behind this work was to evaluate the possible TFA formation after three different extraction methods and application of different conditions of a EF. Previously it was stated that the PPO enzyme is inactivated at 9 kV/cm, 720 Hz and 3 min. In this work the following conditions were applied: voltages of 3 and 9 kV/cm, 60 and 720 Hz, and 10 and 180 s, using a factorial design 23. For this study two strategies were utilized, first the oil was extracted and then treated with the EF, (Oil 1), second the pulp was first treated with the EF and the oil was extracted, (Oil 2). The TFA were analyzed by infrared spectroscopy, (FTIR 400-4000 cm-1) and gas chromatography, (GC). The FTIR results show that all the oils studied had very light amounts of TFA, evidenced by the 968 cm-1 band compared with the very high peaks at 725 cm-1, corresponding to the cis form of the C-H bond. It was also observed that the best method of oil extraction was the centrifugation, (15,557 g at 40 ºC during 10 min) reaching <0.5 g TFA /100g oil, although the yield was very poor, this amount is within the limits of several regulations. In practice, the centrifugated and EF treated avocado oil is safe to the consumer. The traditional extraction method using dissolvent with or without heat had a light increase in the amount of TFA, with 6.1% and 0.7% respectively, compared with centrifugated and EF treated oil. On the chemical indexes of the oils, the oil extracted with dissolvent and heat had a major deterioration. Besides that, when the paste was treated with higher voltage and time of EF the yield of the oil was lightly superior, (6%), however, under these conditions the oils had more TFA. With all these results it is proposed that the EF technology can be used to treat avocado pulp in order to extend their shelf life without apparent losses of the fatty acids and other valuable compounds.
Descripción : Al considerar la demanda mundial de aguacate y sus derivados, y que es este fruto es altamente perecedero, se han buscado nuevas tecnologías para su conservación; el reto principal es disminuir la actividad de la enzima polifenoloxidasa, (PFO), que provoca oscurecimiento de la pulpa del aguacate. En el presente trabajo se evaluó el efecto del campo eléctrico (CE), sobre los ácidos grasos del aguacate cuando se aplica para disminuir la actividad de la enzima PFO. La importancia de esta investigación es que la pulpa de aguacate contiene una importante cantidad de aceite con ácidos grasos funcionales los cuales pueden ser afectados por el CE. Diferentes investigaciones indican que cuando se extraen los aceites por diferentes métodos se generan ácidos grasos trans (AGT). La idea fundamental del presente trabajo fue evaluar la posible formación de AGT en el aceite de aguacate después de someter al aceite y la pasta de este fruto al efecto del CE, obtenidos por tres diferentes métodos de extracción. En el trabajo previo para la inactivación de la enzima PFO en pulpa de aguacate, se encontró que el mejor tratamiento fue con 9 kV/cm a 720 Hz durante 3 min. Para la aplicación del CE en la pulpa y aceite, se utilizaron voltajes de 3 y 9 kV/cm, frecuencias de 60 y 720 Hz, y tiempos de 10 y 180 s, siguiendo un diseño factorial 23. Para el estudio se siguieron dos estrategias: El aceite fue tratado directamente al CE (Aceite 1), en la segunda, la pulpa de aguacate se expuso al CE y posteriormente se extrajo el aceite (Aceite 2). Para el análisis de los AGT se empleó espectroscopia de infrarrojo mediano (FTIR), región de 400- 4000 cm-1, y cromatografía de gases. En los análisis por FTIR en los Aceites 1 y 2 extraídos por los tres métodos propuestos y tratados al CE, se identificó una ligera intensidad en 968 cm-1 relacionada a los dobles enlaces trans, en comparación con la banda a 725 cm-1 que fue más intensa en todas las muestras, y que corresponde a los dobles enlaces cis. El mejor método de extracción fue por centrifugación a 15,557 g a 40 ºC durante 10 min., se cuantificaron isómeros trans <0.5 g/100g, éste límite es el propuesto por la FDA para considerar un aceite como seguro para su consumo. La extracción con disolvente sin y con calor, repercutió en un mayor nivel de isómeros trans con 0.7 y 6.1% respectivamente, más que por la exposición al CE. En cuanto a los índices químicos del aceite obtenido se observó un mayor deterioro por causa del disolvente con calor. Con el incremento del voltaje y tiempo, se cuantificó un aumento en el rendimiento del aceite (6%), sin embargo, con el aumento de estas condiciones se generó ligeramente la formación de AGT en los Aceites 1 y 2. Con los resultados obtenidos, se propone el CE como una alternativa para la conservación de la pulpa de aguacate, ya que mantiene su calidad y otros compuestos de interés como son los ácidos grasos insaturados del aceite de aguacate.
