dc.descriptionLos flavonoides exhiben una fuerte actividad antioxidante y entre otras propiedades son capaces de inhibir el proceso antiinflamatorio por diversos mecanismos de acción, in vitro e in vivo. Investigaciones recientes, en nuestro laboratorio, demostraron la presencia de flavonoides en Eysenhardtia polystachya (Ort.) Sarg. (palo dulce), árbol ampliamente distribuido en México, así como la actividad antiinflamatoria de sus hojas y corteza después de la administración de la infusión y decocción a la dosis de 200 mg/kg de peso corporal (p.c.) y del extracto etanólico a las dosis de 50 y 100 mg/kg de p.c. en un modelo murino de inflamación crónica granulomatosa. Por lo anterior, el objetivo de este estudio fue separar la fracción rica en flavonoides de Eysenhardtia polystachya (Ort.) Sarg. y evaluar su efecto antiinflamatorio, así como su efecto antioxidante. La especie vegetal se recolectó, se identificó, se secó y se molió la corteza. Se preparó un extracto etanólico, a partir del cual se realizó una separación cromatográfica en columna seca. A la infusión y decocción de hojas y corteza, al extracto etanólico así como a las fracciones resultantes de la separación, se les realizó un estudio fitoquímico preliminar y se les cuantificaron los fenoles totales (método de FC), los flavonoides totales (método de quelación con AlCl3) y se les determinó su actividad antioxidante in vitro, al reducir al radical DPPH•. Para evaluar la actividad antiinflamatoria de las fracciones, se utilizaron ratas Wistar hembras adultas de 200 ± 20 g de p.c., a las cuales se les indujo la inflamación utilizando el modelo del granuloma. Se formaron cuatro grupos, el testigo (vehículo, solución de NaHCO3 al 5%, 1 μL/kg de p.c.), el testigo positivo tratado con indometacina (5 mg/kg de p.c.) y los tratados con las fracciones F3. (16-20) y F4. (21-26) a la dosis de 25 mg/kg de p.c. Asimismo, se realizó la disección del estómago y duodeno de las ratas, con la finalidad de observar algún tipo de alteración presente en dichos órganos, posteriormente se les realizó un estudio histopatológico mediante la técnica de H-E. En todos los extractos se detectaron alcaloides, azúcares reductores, cumarinas, glucósidos cardiacos, quinonas, saponinas, taninos y flavonoides (flavonas y flavonoles), pero en el extracto etanólico, la proporción de metabolitos detectados fue mayor con respecto al resto de los extractos. Los pesos secos y finales de los granulomas obtenidos de las ratas tratadas con las fracciones ricas en flavonoides e indometacina disminuyeron con respecto al grupo testigo. Las fracciones no produjeron erosiones, úlceras ni alguna otra alteración en el estómago y el duodeno, en comparación con la que provoca la indometacina. La cantidad de fenoles totales y flavonoides no tuvieron una relación directa con la capacidad antirradical de los extractos, lo cual no quiere decir, que no sean potentes antioxidantes por otros mecanismos o en otros modelos in vitro. Los resultados obtenidos de esta investigación, demuestran que las fracciones ricas en flavonoides inducen un efecto antiinflamatorio probablemente provocado por la inhibición selectiva de la COX-2.es
dc.description.abstractFlavonoids exhibited strong antioxidant activity and other properties that are capable of inhibiting the inflammatory process by different mechanisms of action in vitro and in vivo. Recent research in our laboratory demonstrated the presence of flavonoids in Eysenhardtia polystachya (Ort.) Sarg. (sweet wood) a tree widely distributed in Mexico, as well as anti-inflammatory activity of leaves and bark after the administration of infusion and decoction at a dose of 200 mg/kg (bw) and ethanol extract at doses of 50 and 100 mg/kg (bw) in a murine model of chronic granulomatous inflammation. Therefore, the aim of this study was to separate the flavonoid-rich fraction of Eysenhardtia polystachya (Ort.) Sarg. and evaluate its anti-inflammatory and antioxidant effects. The specie was collected, identified, dried and ground bark. Ethanolic extract was prepared, from which there was a dry column chromatographic separation. The infusion and decoction of leaves and bark, the ethanol extract and the fractions resulting from the separation, were performed a preliminary phytochemical study and were quantified total phenols (FC method), total flavonoids (method of chelation with AlCl3) and their antioxidant activity were determined in vitro by reducing the radical DPPH•. To evaluate the anti-inflammatory activity of the fractions, adult female Wistar rats of 200 ± 20 g (bw) were used, in which inflammation was induced using the model of granuloma. Four groups were formed, the control (vehicle solution NaHCO3 5%, 1 mL/kg, bw), the positive control treated with indomethacin (5 mg/kg, bw) and those treated with fractions F3. (16-20) and F4. (21-26) at a dose of 25 mg/kg, bw. Furthermore, the dissection of the stomach and duodenum of rats was performed, in order to observe any alteration present in these organs, then underwent a histopathological study using the technique of H-E. In all extracts were detected alkaloids, reducing sugars, coumarins, cardiac glycosides, quinones, saponins, tannins and flavonoids (flavones and flavonols), but in the ethanol extract the ratio of metabolites detected was higher compared to the rest of the extracts. Final and dry weights of the granulomas obtained from rats treated with fractions rich in flavonoids and indomethacin significantly decreased as compared to the control group. Fractions did not produce erosions, ulcers or some other alterations in the stomach and duodenum compared with indomethacin. The amount of total phenols and flavonoids were not directly related to the antiradical capacity of extracts, this does not mean, it has not antioxidant activity produced by other mechanisms or detected by other in vitro models. The results of this research show that the fractions rich in flavonoids induce an anti-inflammatory effect in chronic inflammation, probably produced by its selective inhibition of COX-2es
