Título : Efecto del donepecilo y la cerebrolisina en la neuromorfología y actividad motora del ratón

Resumen : Cognitive decline associated with ageing and age-related disorders emerges as one of the greatest health challenges in the next decades. To date, the molecular mechanisms underlying the onset of neuronal physiological changes in the central nervous system (CNS) remain unclear. Studies have indicated the decline in working memory performance in older adults. Similarly, age-related disorders such as Alzheimer´s disease (AD), are associated with changes in the prefontral cortex and related neural circuitry, wich underlines the decline of integrative function between different brain regions. The basal forebrain cholinergic complex comprising medial septum, horizontal and vertical diagonal band of Broca, and nucleus basalis of Meynert provides the mayor cholinergic projections to the cerebral cortex and hippocampus. The cholinergic neurons of this complex have been assumed to undergo moderate degenerative changes during aging, resulting in cholinergic hypofunction that has related to the progressing memory deficits with aging. Then, we evaluated the effect of combination of cholinergic (donepezil) and neurotrophic (cerebrolysin) on the locomotor activity induced by a novel environment as well the effect on the morphology of neuronal cells from the brain of mayor adult male mouse. The DON is a potent and selective acetylcholinesterase inhibitor used in the treatment of Alzheimer´s disease. CBL is a mixture of neuropeptides with effects similar to the endogenous neurotrophic factors and is considered one of the best drugs used in the treatment of dementias such as Alzheimer´s disease. NIH male mouse of 7 months old were obtained, the animals were grouped and each mouse was assigned to either a vehicle (water and “Tween 80” solution 1:4 (1 ml/100 g of body weight), group donepezil DON (0.1 mg/100 g), group cerebrolysin CBL (0.1 g/100 g), and the combined group DON+CBL (0.1 mg/ 0.1 g/100 g), and was administered daily for 9 weeks i.p. After treatment the locomotor activity induced by a novel environment was assessed in vehicle, DON, CBL, and DON+CBL, immediately after the animals were sacrificed and the brains were removed and stained using a modified "Golgi-Cox" method. Finally slices of the brains were obtained and fixed with synthetic resin. Pyramidal cells from layers III and V of the PFC, from BLA, area CA1 of the dorsal and ventral hippocampus, granule cells from the dentate gyrus, and medium spiny neurons of the Shell and core of NAcc were selected for study. We analyzed each neuron and her density distal spines with "Sholl's analysis" (SA) method. We found that the motor activity was increased in the experimental groups compared to control group, nevertheless we did not find the additive effect in the combined therapy. In the prefrontal cortex layer III the DON monotherapy as well as the CBL monotherapy increased the dendritic branching compared to the control group, besides in the combined group (D+CBL) we find pharmacological additive effect demonstrated by the branching increment compared to the control and monotherapy groups. In the distal spines density both monotherapies as well as the combinated therapy increased the spines density compared to the control group, but without showed differences among them. In the layer V of the prefrontal cortex the branching increased in the experimental groups compared to the control group but without showed differences among them, with the exemption in the second and third order dendritic length only with the combined therapy. In dorsal hippocampus CA1 the DON monotherapy did not modify the branching, however the branching decreased in the CBL and therapy combined groups. The dendritic spine density only increased with the DON monotherapy. In the ventral hippocampus CA1, dentate gyrus, basolateral amygdala, and the shell and core of accumbens nucleus the DON and CBL monotherapies increased the dendritic branching of the same way as the combined therapy also increased the branching but without generating additive effects, a similar effect of the treatments on the spines density in these regions were observed. Cerebrolysin was as effective as donepezil, and the combinacion of neurotrophic (CBL) and cholinergic (DON) treatment was effective only in the prefrontal cortex.
Descripción : El declive cognitivo asociado a la edad es uno de los grandes cambios de salud que será de mayor estudio en las próximas décadas. Hasta la fecha los mecanismos moleculares del envejecimiento, respecto a la fisiología neuronal en el sistema nervioso central (SNC) no son claros. Hay estudios señalando que el declive de la memoria de trabajo se presenta en los adultos mayores de la misma manera a los desórdenes de memoria presentados en la enfermedad de Alzheimer (EA); además estos cambios se relacionan con cambios estructurales en los circuítos neuronales que involucran áreas del sistema límbico como la corteza prefrontal (CPF) y el hipocampo, lo cual constituye la base de la degeneración progresiva en las diferentes regiones cerebrales. Es necesaria la investigación de los fármacos que se aplican como tratamiento para EA, el mejor de los casos sería revertir la sintomatología cognitiva. Para este trabajo, elegimos al clorhidrato de donepecilo (DON), y a la cerebrolisina (CBL), el primero actúa como inhibidor reversible de la enzima acetilcolinesterasa (AChE), manteniendo la concentración del neurotransmisor acetilcolina (ACh) en el espacio sináptico y evitando su degradación, mejorando los síntomas cognitivos de la enfermedad. La CBL por su parte, actúa de forma análoga a la de los factores neurotróficos endógenos del cerebro promoviendo la neuroprotección y la neurorregeneración. Con el fin de probar si el tratamiento combinado de DON+CBL ejerce acción farmacológica aditiva, evaluamos en ratón NIH de siete meses los efectos de la administración intraperitoneal i.p. durante nueve semanas de CBL, DON, y su combinación DON+CBL sobre: La actividad motora y la morfología de neuronas piramidales de las regiones CA1 ventral y dorsal del hipocampo, células granulosas del giro dentado (GD), las piramidales de la corteza prefrontal capas III y V, las piramidales de la amígdala basolateral (ABL), así como las neuronas espinosas medianas del centro y corteza del núcleo accumbens (NAcc). Se formaron cuatro grupos experimentales tratados durante nueve semanas con inyección i.p. diaria de solución vehículo (VEH): Tween 80+agua en proporción 1:4 (1 mL/100 g de peso del animal), grupo DON (0.1 mg/100 g), grupo CBL (0.1 g/100 g), y DON+CBL (DON 0.1 mg/ CBL 0.1 g). Después del tratamiento, se realizó la prueba conductual de “actividad motora en ambiente nuevo y campo cerrado”, posteriormente se sacrificaron los animales y se extrajeron los cerebros a continuación se trataron con la tinción de “Golgi-Cox” e inmediatamente se prosiguió a realizar los cortes histológicos y revelado de los mismos, finalmente se fijaron los cortes a portaobjetos con resina sintética, y se prosiguió a la representación bidimensional de las neuronas y sus espinas, por medio de un microscopio óptico adaptado a una cámara lúcida. Por último analizamos cada neurona obtenida así como sus espinas distales aplicando la técnica del “Análisis de Sholl” (AS) y evaluando así los parámetros de arborización, longitud por orden dendrítico, y longitud total dendrítica. Los resultados fueron los siguientes: la actividad motora aumentó en los tres grupos experimentales frente al control (incluyendo al grupo de terapia combinada), sin embargo, no encontramos el efecto aditivo en el tratamiento combinado. Para la corteza media prefrontal (CmPF) capa III, tanto la monoterapia de DON, como la de CBL, aumentaron la arborización dendrítica respecto al control, además de que, en el grupo combinado (DON+CBL) obtuvimos efecto farmacológico aditivo, visto por el aumento de la arborización frente al control y los grupos con monoterapia; respecto a la densidad de espinas distales, tanto las monoterapias como la combinación de ambas drogas, aumentaron la densidad de espinas frente al control, pero sin haber diferencias entre ellas. En la capa V de la corteza media prefrontal, la arborización aumentó en los tres grupos experimentales frente al control, pero sin haber diferencias entre ellos, a excepción de que, en el segundo y tercer orden, aumentó la longitud dendrítica sólo con la terapia combinada. En hipocampo dorsal CA1, la monoterapia con DON no modificó la arborización, pero tanto la de CBL como la terapia combinada de ambas drogas, disminuyó la arborización. Con respecto a la densidad de espinas dendríticas, ésta sólo aumentó con DON. Respecto a la región CA1 del hipocampo ventral, giro dentado, amígadala basolateral, y el núcleo accumbens; tanto la monoterapia de DON como la de CBL aumentaron la arborización, de la misma forma, la terapia combinada de ambas drogas aumentó la arborización, pero sin generar efecto sumatorio, el efecto sobre la densidad de espinas fue el mismo. Por lo tanto, concluimos que el tratamiento con ambas drogas generó efecto aditivo sólo en la corteza prefrontal.
