Título : EVALUACIÓN DE LA ACTIVIDAD DE LA ENZIMA ARGINASA DURANTE LA PREECLAMPSIA

Resumen : Preeclampsia is the leading cause of maternal-fetal, characterized by hypertension and proteinuria. It has been reported that hypertension is due to the low availability of nitric oxide produced by endothelial dysfunction, oxidative stress and uncoupling of nitric oxide synthase (NOS). We propose that another possible event linked to low availability of NO• is the increased activity of the enzyme arginase, which is an enzyme that competes with nitric oxide synthase (NOS) for the substrate L-arginine, as shown by reports from other diseases with hypertension. The aim of this study was to evaluate the activity of the enzyme arginase and its relationship to oxidative stress. This involved four groups of women: 1) Healthy volunteer (HV) with Body Mass Index (BMI) of 18.5 to 24.9 Kg/m2, 2) Obese 1 (O1) with BMI of 30 to 34.9 (grade 1), 3) Normal pregnant (NP) women and 4) Pregnant women with preeclampsia (PE). Group 3 and 4 pregnancy week 20 to 40. The PE group was caught in the emergency room with blood pressure of 140/90 mmHg and proteinuria of 0.3 g of protein in urine for 24 hours in 2 mediations with an interval of 6 hours. The ages of all participants were between 20 and 30 years without any treatment. They provided a blood sample for plasma, which determines the arginase enzyme activity and damaged was also determined by oxidative stress to lipids by the method of 1-methyl-2-phenylindole and protein quantifying carbonyl groups as a marker of antioxidant defense total SH groups were quantified, also was realized the quantification of nitrite/nitrate. To analyze the results we used ANOVA with post hoc test of Bonferroni. The results were expressed as mean ± EE and were considered significant at p< 0.05. Where the results showed that all PE patients were obese compared with NP. We also detected that the O1 and PE patients showed similar values for arginase activity, less activity was found in the O1 and PE (0.30 ± 0.04 and 0.31 ± 0.04 nmol/mg protein/minute, respectively) than in HV and NP (0.42 ± 0.04 and 0.45 ± 0.05, respectively). From Interesting way, there was a similar pattern in the total SH group (3.94 ± 0.71 and 4.29 ± 0.39 nmol/mg protein, respectively) however, these values were lower than HV and NP (6.19 ± 0.37 and 5.91 ± 0.43, respectively), which may be due to oxidative characteristics of obesity that preeclamptic patients enrolled. This decrease in SH concentration could be due to its nitration by peroxynitrite resulting nitrosothiols. Taken together our results support the role of oxidative stress in preeclampsia and its close relationship with obesity. So, this work opens the possibility of creating preventive strategies, It's for this, that more studies are needed in this subject.
Descripción : La Preeclampsia es la principal causa de mortalidad materno-fetal, caracterizada por hipertensión y proteinuria. Se ha descrito que la hipertensión es debida a la baja disponibilidad del oxido nítrico producido por la disfunción endotelial, estrés oxidativo y desacople de la oxido nítrico sintasa (NOS). Nosotros proponemos que otro posible evento ligado a baja disponibilidad del NO• es la actividad incrementada de la enzima arginasa; la cual, es una enzima que compite con la sintasa de óxido nítrico (NOS) por el sustrato L-arginina, como lo han mostrado reportes de otras enfermedades que cursan con hipertensión. El objetivo de este trabajo fue evaluar la actividad de la enzima arginasa y su relación con el estrés oxidativo. Para ello participaron 4 grupos de mujeres: 1) voluntarias sanas (VS) con índice de masa corporal (IMC) de 18.5 a 24.9 Kg/m2; 2) con obesidad grado 1 (O1) con IMC 30 a 34.9; 3) con embarazo normal (EN); y 4) con preeclampsia (PE). El grupo 3 y 4 con embarazo de la semana 20 a la 40. El grupo PE fue captado en urgencias con presión arterial de 140/90 mmHg y proteinuria de 0.3 g de proteína en orina de 24 horas en2 mediaciones con intervalo de 6 horas. Las edades de todas las participantes fueron entre 20 a 30 años sin ningún tratamiento. Se les tomó una muestra de sangre para obtener plasma, donde se determinó la actividad de enzima arginasa, así mismo se determinó daño por estrés oxidativo a lípidos mediante el método de 1-methyl-2-phenylindole y a proteínas cuantificando grupos carbonilos y como marcador de defensa antioxidante se cuantificaron grupos SH totales; además se realizó la cuantificación de nitritos/nitratos. Para analizar los resultados se utilizó ANOVA con una prueba pos hoc de Bonferroni. Los resultados fueron expresados como promedios +/- EE y se consideró significativo una p<0.05. Donde resultó que todas las pacientes PE tenían obesidad en comparación a las de EN. También detectamos que las pacientes O1 y PE mostraron valores similares en la actividad de arginasa, la menor actividad se encontró en las pacientes O1 y PE (0.30 ± 0.04 y 0.31 ± 0.04 nmol/mg proteína/minuto, respectivamente) en comparación con los grupos VS y EN (0.42 ± 0.04 y 0.45 ± 0.05, respectivamente). Interesantemente, hubo un comportamiento similar en los grupos SH (3.94 ± 0.71 y 4.29 ± 0.39 nmol/mg proteína, respectivamente). Sin embargo, fueron valores menores a los grupos de VS y mujeres con EN (6.19 ± 0.37 y 5.91 ± 0.43, respectivamente), lo cual puede ser debido a las características oxidativas de la obesidad por las que cursan las pacientes preeclampticas. Esta disminución de las concentraciones de SH puede ser debida a la nitración por parte de peroxinitrito, dando como resultado nitrosotioles.
